3 Figure 2 Alignment of dmrt1-Related Sequences of the Genus Oryzias and Phylogenetic Analysis A coding region of 693 bp corresponding to 231 amino acids of Ocedmrt1 was obtained. (A) Amino acid sequences of dmrt1-related genes were deduced and aligned with Clustal W. Carbio and HNI are two O. latipes strains belonging to the southern and northern Japanese populations, respectively. The dark and light shaded boxes indicate amino acids conserved among the five sequences. The lines over the sequences indicate the DM domain. The amino acid sequences at the beginning and the end of Ocedmrt1 were not determined because PCR primers for cloning were designed in those regions. (A and B) Genomic DNAs from male (M) and female (F) O. celebensis were digested with EcoRI and hybridized with Oladmrt1a (A) and Oladmrt3 (B). (C and D) Male and female genomic DNAs of O. latipes (strain Carbio) digested with BamHI (C) and O. celebensis digested with HindIII, PstI, and PvuII (D) were hybridized with OlaFLNK. Note that additional male-specific bands are present in O. latipes (C), representing the duplication of this sequence on the Y chromosome. The hybridization conditions in (A), (B), and (D) were moderate stringency, and in (C) they were high stringency. (C and D) PstI-digested female (genotype: XX) and male (genotype: XY) tilapia DNAs were hybridized with HsaDMRT1 (C) and rehybridized with Oladmrt1a (D). (E) PstI- or HindIII-digested O. mekongensis DNAs were hybridized with Oladmrt1a probe. (F) Hybridization of EcoRI-digested O. latipes (strain Carbio) genomic DNAs with HsaDMRT1 reveals a male-specific band (arrow). The hybridization conditions were moderate stringency.
