Leukemic cell populations were purified from host T cells via the unique Vβ receptor. (B) ATAC-seq on each population revealed profiles of active cis-regulatory elements, with evidence of population-distinct gene signatures, unique transcription factor circuits associated with response or resistance to HDACi, and correlations with subsets of memory T cells. (C) Chromatin states during HDACi therapy were assessed, revealing that response is associated with pharmacodynamic increase in baseline regulatory element accessibility and apparent normalization of host T cell chromatin signatures.
