Supplementary Figure1. Systemic administration of LPS does not lead to a general induction of EGFP+ expression in CD11b+ microglia in different regions of the brain. Lys-EGFP-ki mice (n=4) were injected intravenously with LPS (4.5 μg/g). 24 h later the mice were euthanized, perfused with 4% paraformaldehyde in PBS and their brains were sectioned on a cryostat. Sections (16 μm) stained with anti-EGFP (green fluorescence) and anti-CD11b (red fluorescence). Representative digital confocal images from the midline (1st row) and neocortex (2nd row) of the brain are shown in brightfield (DIC, first column) and with fluorescence for EGFP (2nd column) and CD11b (3rd column). A merged image is also shown (4th column). Control mice not injected with LPS (3rd row) or not stained with the CD11b primary antibody (4th row) are also shown. EGFP+CD11b+ cells are found only in the pia or in blood vessels (bv) but not in CD11b+ microglia. Scale bar = 50 μm.
