B7-H4 expression correlates with MHC-I expression and improved prognosis in patients with breast cancer. B7-H4 expression correlates with MHC-I expression and improved prognosis in patients with breast cancer. A, histologic sections of mammary tumors from WAP-gp/MMTV-PyMT (n = 6) and WAP-gp/MMTV-PyMT/B7-H4−/− (n = 5) mice were taken 15 days after LCMV infection and stained with an anti–MHC I antibody. B, single-cell suspensions from human breast tumor samples (n = 20) were stained for CD45, B7-H4 and HLA-A, -B, -C. A Spearman correlation analysis was performed. C, SKB-R3, MCF-7 and HCC-1143 cells untreated or transfected with either B7-H4-specific siRNA or control, scrambled siRNA. At 48 hours after transfection, cell-surface expression of B7-H4 and HLA-A, -B, -C was determined by flow cytometry. Data shown are representative of three independent experiments. D, the Kaplan–Meier plot of recurrence-free survival for 2,681 patients with breast cancer, stratified by high versus low B7-H4 gene expression relative to the median value of the entire dataset. *, P < 0.05.
