Fig FTY720 suppresses experimental abdominal aortic aneurysms (AAAs). Aneurysms were created via intra-aortic porcine pancreatic elastase (PPE) infusion in 10-week-old male C57BL/6J mice, which were daily treated with FYT720 (A-G) 3 days before (1 mg/k) or (H) 4 days after (0.3 and 1 mg/kg), PPE infusion. Influences on aneurysm formation and progression were evaluated via ultrasonography and histology. A, Ultrasound-measured aortic diameters before and after PPE infusion. Scores for (B) medial elastin degradation (Verhoeff staining) and (C) smooth muscle cell (SMC) destruction (SMC-α actin staining). Quantification of (D) CD68+ macrophages, (E) CD3+ T cells, (F) B220+ B cells, and (G) CD31+ blood vessels in aortic sections. H, Changes in aortic diameters in mice with existing AAAs after FTY720 or vehicle treatment. ACS, Aortic cross-section. All data are mean and standard deviation from six to seven mice in each group. *P < .05 and **P < .01 compared with vehicle treatment.
