2 Figure 1 Expression of visfatin messenger RNA and protein in the ovaries of differently aged mice. Whole ovaries were collected at the indicated time after PMSG injection. (A, B) Representative RT-PCR (A) and Western blot (B) for visfatin expression of 6- and 26-week-old mice. Visfatin mRNA and protein intensity values were normalized to those of GAPDH and β-actin, respectively, and were expressed as mean ± SD. N = 3, six ovaries per age group. #P<.001 vs. control (0 hour). Fertility and Sterility  , e3DOI: ( /j.fertnstert ) Copyright © 2012 American Society for Reproductive Medicine Terms and Conditions

4 Figure 3 Effect of coadministration of visfatin on expression of ovarian visfatin and VEGF in differently aged mice. Various doses (0–500 ng/mL) of recombinant mouse visfatin were administered intraperitoneally to two groups of mice (6–11 weeks old and 26–31 weeks old) during superovulation with PMSG and followed by hCG approximately 48 hours later. Then the mice were immediately paired with an individual male. Eighteen hours after hCG injection both ovaries of each mouse were collected, and the expressions of ovarian visfatin and VEGF was examined by RT-PCR. (A) Representative RT-PCR for ovarian visfatin expression of 6- and 26-week-old mice. (B) Visfatin messenger RNA intensity values were normalized to those of GAPDH and expressed as mean ± SD. N = 3, six ovaries per age group. #P<.05, ∗P<.001 vs. control (0 ng/mL). (B, H) Twenty-six-week-old-mice. (C, I) Visfatin at 5 ng/mL. (D, J) Visfatin at 10 ng/mL. (E, K) Visfatin at 100 ng/mL. (F, L) Visfatin at 500 ng/mL. Magnification, ×100.
