1
Creating a DNA Fingerprint by Gel Electrophoresis
2
Gel Electrophoresis Method for separating molecules (DNA, proteins, etc.) on the basis of physical or chemical properties such as: (1) size (2) shape (3) electrical charge
3
Overview of Equipment Gel electrophoresis apparatus - An agarose gel is placed in this buffer-filled box and electrical field is applied via the power supply to the rear. The negative terminal is at the far end (black wire), so DNA migrates toward the camera.
4
Electrophoresis of DNA
Gels are made of agarose or polyacrylamide DNA samples loaded, voltage applied Negatively charged DNA migrates toward “+” electrode Smaller DNA fragments migrate faster
5
Agarose powder (mix with liquid and heat to make gel)
Place comb into gel before it hardens, to make wells for DNA Use micropipette with clean tip to inject DNA sample into well
