Figure 2 Protease activity, active MMP9 and ADAMTS-5. Bovine FDCex were pre-cultured for 1 week, and then cultured for further 5 weeks with weekly administration of indicated compounds. MMP9 (A) and ADAMTS-5 (B) (Nordic Bioscience, Denmark) were measured in conditioned medium collected at 0,1, 2, 3, 4 and 5 weeks of compound-culturing. Values are means±SEM of six replicate FDCex after normalization of individual FDCex to their baseline value. One-way ANOVA was used to compare each point with its respective vehicle group. Significance levels are indicated by asterisks; *P < 0.05, **P < 0.01, ***P < 0.001 Osteoarthritis and Cartilage , S157-S158DOI: ( /j.joca ) Copyright © Terms and Conditions

Figure 1. Type II collagen and aggrecan degradation, C2M, AGNx2 and AGNx1. Bovine FDCex were pre-cultured for 1 week, and then cultured for further 5 weeks with weekly administration of indicated compounds. C2M (A), AGNx2 (B) and AGNx1 (C) (Nordic Bioscience, Denmark) were measured in conditioned medium collected at 0, 1, 2, 3, 4 and 5 weeks of compound-culturing. Results from two studies on two different cows (Upper pane, Cow#1; Lower pane, Cow#2) are shown. Values are means±SEM of six replicate FDCex after normalization of individual FDCex to their baseline value. One-way ANOVA was used to compare each point with its respective vehicle group. Significance levels are indicated by asterisks; *P < 0.05, **P < 0.01, ***P < Osteoarthritis and Cartilage , S157-S158DOI: ( /j.joca ) Copyright © Terms and Conditions
