1 A Recombinant Probiotic for Treatment and Prevention of Cholera
Antonio Focareta, James C. Paton, Renato Morona, Jan Cook, Adrienne W. Paton  Gastroenterology  Volume 130, Issue 6, Pages (May 2006) DOI: /j.gastro Copyright © 2006 American Gastroenterological Association Institute Terms and Conditions

2 Figure 1 Predicted oligosaccharide structure of the LPS of E coli CWG308:pGM1a/pCst. The portion present in the LPS of E coli CWG308 (based on published studies6) is shown in normal font, whereas the GM1-mimic portion synthesised by the heterologous glycosyltransferases is shown in bold. Linkages formed by the various transferases are identified with dotted arrows. Sugars are abbreviated as follows: Gal, galactose; Glc, glucose; GlcNAc, N-acetylglucosamine; GalNAc, N-acetylgalactosamine; NeuAc, N-acetylneuraminic acid; Hep, l-glycero-D-manno-heptose; Kdo, 3-deoxy-D-manno-oct-2-ulosonic acid. 3 Figure 2 (A) Silver-stained PAGE analysis of LPS from E coli CWG308 and derivatives carrying pGM1a/pCst, pGb3, and pGb4 (the latter 2 plasmids result in addition of 2 and 3 sugars to the CWG308 LPS core, respectively). LPS of CWG308 and CWG308:pGM1a/pCst separated by PAGE was also blotted onto PVDF, probed with either polyclonal anti-GM1 (B), or peroxidase-conjugated CtxB (C) and developed as described in the Materials and Methods section. 4 Figure 3 Binding of peroxidase-conjugated CtxB to CWG308 and CWG308:pGM1a/pCst cells. Serial 4-fold dilutions of indicated bacterial suspensions in PBS were spotted onto nitrocellulose and reacted with peroxidase-conjugated CtxB, as described in the Materials and Methods section. 5 Figure 4 Effect of adsorption with CWG308:pGM1a/pCst on cytotoxicity of Ctx for Y1 cells. Aliquots of Ctx (2.5 μg) were preabsorbed with CWG308 or CWG308:pGM1a/pCst cells suspended in PBS or PBS alone. Cells were removed by centrifugation, and filter-sterilized supernatants were then diluted 80-fold, and 50 μL was added to Y1 monolayers, followed by 150 μL of DMEM (Materials and Methods section). Monolayers were photographed under phase-contrast microscopy (×20 objective) after 6-hour incubation.
