Accumulation of fumarate, succinate, or D-2HG by mutation of the related enzymes leads to inhibition of TET demethylases, among other enzymes. This leads to the decreased expression of the miR-200 family and E-cadherin and an increase in Twist1 and vimentin, among other target genes. Snail1 and Snail2 also induce EMT by repression of critical genes such as E-cadherin.
