8 Method for Measuring Rosé Color Components
James F. Harbertson Washington State University Viticulture and Enology Program General This is a simple assay designed to measure the components of color in rosé wine. The assay measures the color of the rosé wine, the amount of color that is bleached by sulfur dioxide and the amount of color that is polymeric in nature (will not bleach with the addition of sulfur dioxide). The analysis only requires a simple spectrophotometer capable of measuring absorbance at 520nm and simple cuvettes that are transparent in the visible spectra. Samples do not require pH adjustment as the analysis is designed to be a measure of the wine in its current state. The analysis is quick and simple and is designed to provide relative values for winemakers to make decisions during wine production.

Harbertson Washington State University Viticulture and Enology Program Calculations: Color of Rose Wine [C] = A520 Color due to Bleached Anthocyanin [BA] = (Aacet – A520) absorbance units Total Potential Color of Rose Wine [TC] = BA + C Color due to Polymeric Pigment [P] = 1.08 * ASO2 absorbance units Measurements A520nm: 1 mL of wine sample is added to a 10 mm plastic cuvette. Measure absorbance at 520 nm. Aacet: 10 μL of 10% acetaldehyde solution is added to 1 mL of wine sample in a 10 mm plastic cuvette. Vortex cuvettes gently to ensure homogenization. After 45 minutes, the sample is measured at 520 nm. ASO2: 80 μL of 5% SO2 solution is added to 1 mL of wine sample in a 10 mm cuvette. Vortex cuvettes gently to ensure homogenization. After 10-min incubation, the absorbance is measured at 520 nm.
