1 Association Between Obesity and Discordance in Fibrosis Stage Determination by Magnetic Resonance vs Transient Elastography in Patients With Nonalcoholic Liver Disease  Cyrielle Caussy, Jun Chen, Mosab H. Alquiraish, Sandra Cepin, Phirum Nguyen, Carolyn Hernandez, Meng Yin, Ricki Bettencourt, Edward R. Cachay, Saumya Jayakumar, Lynda Fortney, Jonathan Hooker, Ethan Sy, Mark A. Valasek, Emily Rizo, Lisa Richards, David A. Brenner, Claude B. Sirlin, Richard L. Ehman, Rohit Loomba  Clinical Gastroenterology and Hepatology  Volume 16, Issue 12, Pages e7 (December 2018) DOI: /j.cgh Copyright © 2018 AGA Institute Terms and Conditions

2 Figure 1 Concordance and discordance-rate between MRE and TE for the stage of liver fibrosis. Concordance and discordance-rate are presented as percentage of the total cohort in (A) the USCD NAFLD Cohort (N = 119) and (B) the Mayo Clinic NAFLD Cohort (N = 75). 3 Figure 2 Discordance-rate between MRE and TE increases as the BMI increases. Discordance-rate between MRE and TE for the stage of liver fibrosis (2–4 vs 0–1) represented in percentages of individuals across BMI category in (A) the UCSD NAFLD Cohort and (B) the Mayo Clinic NAFLD Cohort. *P value for trend determined using a Cochran-Armitage test for trend. 4 Figure 3 Discordancy between MRE and TE is significantly higher in severely obese individuals. Concordance and discordance-rate between MRE and TE for the stage of liver fibrosis (2–4 vs 0–1) as presented as percentage of (A) individuals with BMI <35 kg/m2 (n = 92) compared with individuals with BMI ≥35 kg/m2 (n = 27) in the UCSD Cohort and (B) nonmorbid obese (n = 28) compared with morbid obese (n = 47) individuals. P value determined using a Pearson chi-square test. Clinical Gastroenterology and Hepatology  , e7DOI: ( /j.cgh ) Copyright © 2018 AGA Institute Terms and Conditions

5 Supplementary Figure 1 Mayo Clinic NAFLD cohort flow-chart. Among the 111 subjects from a previously published study,13 a total of 19 were excluded because of other etiology than NAFLD and 92 subjects with biopsy-proven NAFLD were eligible. Of these 92 subjects, 10 were excluded because TE was not available and 7 were excluded because MRE was not available. A total of 75 subjects were included in the final analysis. 6 Supplementary Figure 2 Distribution of MRE and TE measurements in concordant and discordant group. Distribution of MRE and TE measurements in the UCSD NAFLD Cohort and Mayo Clinic NAFLD Cohort among (A) MRE and TE concordant group and (B) MRE and TE discordant group. 7 Supplementary Figure 3 Distribution of concordance and discordancy between MRE and TE. (A) In UCSD NAFLD Cohort, the discordance-rate was 51.2% (n = 62). Among the discordant group, 51.9% (n = 27) occurred when only TE was discordant, 21% (n = 14) occurred when only MRE was discordant, and 21% (n = 11) occurred when both MRE and TE were discordant. (B) In the Mayo Clinic NAFLD Cohort, the discordance-rate was 54.2% (n = 34). Among the discordant group, 58.8% (n = 20) of the discordancy occurred when only TE was discordant, 14.7% (n = 5) occurred when only MRE was discordant, and 26.4% (n = 9) occurred when both MRE and TE were discordant. 8 Supplementary Figure 4 Distribution of BMI across concordant and discordant group. (A) In the UCSD NAFLD Cohort, the mean BMI when both MRE and TE were discordant and when only TE was discordant were significantly higher compared with the concordant group (34.2 kg/m2 and 32.2 kg/m2 vs 29.5 kg/m2; P = .003 and P = .02, respectively). (B) In the Mayo Clinic NAFLD Cohort, the mean BMI when only TE was discordant was significantly higher compared with the concordant group (45.0 kg/m2 vs 39.9 kg/m2; P = .011). *P value <.05 determined using independent sample Student t test compared with concordant group.
