Figure 1 Blastocyst expansion and hourly interval rates of Change in known positive implantation outcomes (KID+) and known negative implanatation outcomes (KID−) human embryos; (a) the average cross sectional area (CSA) in 100% KID+ (black) and KID− (red) blastocysts up to 19 h from blastocyst formation. Each group shows continuous expansion although there is an inflection point at 12 and 15 h in the KID+ and KID− groups, respectively. Error bars reflect SEM; (1B) averaged hourly interval rate of change (IRC) in cross-sectional area in KID+ (black) and KID− (red) blastocysts. Each demonstrates an underlying pulse-like or oscillatory nature during expansion. Reproductive BioMedicine Online , DOI: ( /j.rbmo ) Copyright © Terms and Conditions

Figure 2 Blastocyst expansion and hourly interval of change (IRC) in cross-sectional area in two known positive implantation outcomes (KID+) embryos; (2A) this embryo was monitored for 11 h and shows continuous expansion through E-type (expansion) oscillations (positive IRC values) to over 25,000 µm2. There are no C-type contractions; (2B) this embryo was monitored for 16 h. Exhibiting E-type (expansion) oscillations, it shows a continuous rate of expansion to over 25,000 µm2 at 12 h but then demonstrates a single C-type (contraction) oscillation, followed by rapid recovery and further expansion to nearly 30,000 µm2 at 16 h. CSA, cross-sectional area. Reproductive BioMedicine Online , DOI: ( /j.rbmo ) Copyright © Terms and Conditions

Figure 3 Morpokinetics of recovery after C-type contraction in a human known positive implantation outcome (KID+) embryo; (a) appearance of human KID+ blastocyst before collapse at 97.2 h after intracytoplasmic sperm injection (left), after collapse at 97.8 h (centre), and after recovery at 99.6 h (right). Scale bar = 100 microns; (3B) kinetics of the collapse and recovery. Cross-sectional area (CSA) and hourly interval rate of change (IRC) measurements from sequential frame analysis at 0.2–0.3 h intervals. The loss of area is rapid and followed by a more gradual recovery over 2–3 h. The interval rate of change plot reveals acceleration of expansion during blastocoel volume recapture. Figure 4 Regression analysis of blastocoel expansion as a function of the time of blastocyst formation after intracytoplasmic sperm injection. Comparison of the cross-sectional area (CSA) of known positive implantation outcome (KID+) embryos (black) and known negative implantation outcome (KID−) embryos (red) at 3 h (A), 9 h (B), and 12 h (C) from time of blastocyst formation. At 3 and 9 h, the regression lines (a, KID− and b, KID+) have negative slopes only for the KID− group. At 12 h, the regression line slopes are positive for each group (Table 2).
