Investigations of the Binding of Peripheral Membrane Proteins
J.J. Breen, Dept. of Chemistry & Biochemistry, Providence College, Providence, RI 02918 Project Aim: To test the extended lipid conformation hypothesis. Fluorescence for pyrene labeled vesicles is quenched upon the addition of cyt c. Subsequent addition of unlabeled DOPG lipid in a large increase in the fluorescence signal for labeled vesicles containing N-acylated DPPE lipids (C16>C2) in contrast to that observed for labeled vesicles containing DOPG. The cyt c quenched fluorescence of pyrene in labeled PG vesicles increases upon the addition of unlabeled DOPG vesicles to a increasingly greater extent as the acyl chains of the PG lipids in the labeled vesicles decrease in length (C18C8). vs.
