2 Figure 1 FL and IL-12 differentially regulate DC in murine epidermis and dermis. C57BL/6 mice were injected i.p. with 10 μg FL, or 1 μg IL-12, both together, or 0.01% mouse serum albumin for 10 consecutive days. Animals were sacrificed 24 h after the last injection. Histograms represent the number of DC per mm2 (y-axis) in epidermis (solid bars) and dermis (stippled bars) in control mice and after cytokine treatment (x-axis). Upper panel: NLDC-145+ cells in wild-type (a) and IL-12 knockout mice (b). Lower panel: CD86+ cells in wild-type (c) and IL-12 knockout mice (d). The means ± SEM from three separate experiments are shown. *p < 0.05 compared with the control by one-way ANOVA. 3 Figure 2 Effect of FL treatment on the expression of NLDC-145 and CD86 in wild-type mice. (a, b) NLDC-145; (c, d) CD86. Immunohistochemistry shown is representative of three independent experiments. (a) Control, (b) FL treatment. FL treatment resulted in a significant accumulation of both NLDC-145+ cells (b) and CD86+ cells (d) preferentially in the dermis compared with the control specimens (a, c). Scale bar: 110 μm. 4 Figure 3 IL-12 significantly decreased and FL slightly increased the epidermal thickness. Histogram showing the epidermal thickness in μm (y-axis) in different treatment groups (x-axis). Solid bars represent the data from wild-type mice and stippled bars represent the data from IL-12 knockout mice. Histograms show the means (± SEM) based on three independent experiments. *p < 0.05 vs control by one-way ANOVA. 5 Figure 4 FL and IL-12 reciprocally change epidermal cellularity. Histogram showing epidermal cellularity in cells per mm2 (y-axis) in different treatment groups (x-axis). Solid bars represent the data from wild-type mice and stippled bars represent the data from IL-12 knockout mice. In wild-type mice, FL administration induced an increase in epidermal cell numbers whereas IL-12 induced a decrease. In IL-12 knockout mice, we observed the opposite effect. Histograms show the means ± SEM based on three independent experiments. *p < 0.05 vs control by one-way ANOVA. 6 Figure 5 FL induces an increase in dermal cellularity. Histogram showing dermal cellularity in cells per mm2 (y-axis) in different treatment groups (x-axis). Solid bars represent the data from wild-type mice and stippled bars represent the data from IL-12 knockout mice. Histograms show the mean (±SEM) based on three independent experiments. *p < 0.05 vs control by one-way ANOVA.
