Hep G2 cells were seeded in flasks (1 × 106 cells/mL) and exposed to either plain medium (lane A) or 80 mmol/L EtOH for 24 hours (lane B) or to 80 mmol/L EtOH for two 24-hour periods (lane C). Total cellular RNA was isolated from Hep G2 cells. Cytokine gene expression was evaluated by RT-PCR. The amount of DNA was normalized for the amount of G3PDH. Analysis of the expression was performed in triplicate in at least three different cell populations. Data are given as percentages with controls at 100%. The PCR product was electrophoresed on a 2% agarose gel. The expression of each specific cytokine was quantitated by densitometry and is given as a percentage, with controls at 100%.
