Figure S1 Linezolid, vancomycin and daptomycin differentially modulate the expression of cytokines in monocytes. THP-1 monocytes were incubated with linezolid (LIN), vancomycin (VAN) or daptomycin (DAP) in the presence or absence of LPS for 2 h or 6 h. Quantitative Real-Time-PCR (qRT-PCR) analysis was performed to determine the expression levels of genes encoding TNF (A,B), IL-1β (C,D), IL-6 (E,F) and IL-10 (G,H) after normalization to GAPDH. Each bar represents the fold-change in mRNA expression comparing unstimulated control with antibiotic-incubated cells or LPS-stimulated control with LPS plus antibiotic-stimulated cells, respectively. Data are expressed as mean ± SEM of six independent experiments; *, p < 0.05 vs. respective LPS-free control; +, p < 0.05 vs. respective LPS control. Figure S2 Linezolid, vancomycin and daptomycin differentially modulate the expression of TLRs in monocytes. THP-1 monocytes were incubated with linezolid (LIN), vancomycin (VAN) or daptomycin (DAP) in the presence or absence of LPS for 2 h or 6 h. Quantitative Real-Time-PCR (qRT-PCR) analysis was performed to determine the expression levels of genes encoding TLR1 (A,B), TLR2 (C,D), TLR4 (E,F), TLR6 (G,H), TLR7 (I,J) and TLR9 (K,L) after normalization to GAPDH. Each bar represents the fold-change in mRNA expression comparing unstimulated control with antibiotic-incubated cells or LPS-stimulated control with LPS plus antibiotic-stimulated cells, respectively. Data are expressed as mean ± SEM of six independent experiments; *, p < 0.05 vs. respective LPS-free control; +, p < 0.05 vs. respective LPS control.
