Figure 1 Regulation of Epidermal Homeostasis through Dnmt3 Regulation of Enhancers Dnmt3a and Dnmt3b associate with active enhancers in epidermal stem cells to potentially control the expression of genes involved in regulating stem cells. In the case of Dnmt3a, it binds to the center of enhancers along with the transcription factor p63 and Tet2. Tet2 catalyzes the conversion of 5-methylcytosine (5-mc) to 5-hydroxymethylcytosine (5-hmc). With Dnmt3b, it binds broadly throughout the enhancer region through association with the H3K36me3 mark. During differentiation the expression of Dnmt3b is diminished while the expression of Dnmt3a is increased. This allows Dnmt3a to bind to enhancers that potentially regulate differentiation genes.
