Activation of heterotrimeric G proteins generates free Gαq/12/13 and Gβγ subunits, which mediate the membrane recruitment and activation of Csk. The Gβγ subunit effect involves the physical association of Gβγ and Csk, while the Gαq/12/13 effect is mediated through as yet unknown intermediates. Csk activates Rho either by relieving the Rho inhibition that is caused by Src-mediated activation of p190RhoGAP or by alternative mechanisms, possibly including activation of a RhoGEF.
