3 Figure 1 Phylogenetic Tree of Plant and Prokaryotic Multidrug and Toxic Compound Extrusion (MATE) Family of Proteins The amino acid alignment of 32 plant proteins and 20 prokaryotic proteins of the MATE family and the MATE protein from Camelina sativa (CasMATE) was obtained with MEGA7. The NCBI Reference Sequence ID of the MATE proteins are listed in Table S1. The phylogenetic tree data were calculated with the maximum likelihood method using MEGA7. The tree illustration was obtained using Figtree ( Groups A, B, C, and F correspond to plant MATE proteins. CasMATE is underlined in red. Groups D and E correspond to prokaryote MATE proteins. PDB IDs are shown in parentheses. See also Figure S1 and Table S1. Structure  , e2DOI: ( /j.str ) Copyright © 2017 Elsevier Ltd Terms and Conditions

4 Figure 2 Overall Structure of CasMATE
(A) Ribbon representation of CasMATE viewed from the membrane region and extracellular sides. The number of transmembrane helices (TM) are shown. Each helix is colored blue (N terminus) to red (C terminus). (B) Close-up views of the N lobe and C lobe of the internal pocket. The side chains of glycine, proline, phenylalanine, tryptophan, aspartic acid, glutamic acid, and asparagine residues in the pocket surface are shown as stick representations. See also Figure S2. 5 Figure 3 Polarity of the Internal Pocket in CasMATE
(A and B) Cross-section model of CasMATE surface representation is shown from the membrane region (A) or from the extracellular (B) sides. Hydrophilic residues are shown as stick representations. (C) Pocket polarity of the reported MATE structures (PDB: 3MTK, 4HUM, 3VVN, 5C6N, and 4Z3N) viewed from the same angles as in (A) and (B). The electrostatic potentials calculated by APBS and PDB2PQR packages (Dolinsky et al., 2004) are colored ranging from blue (+20 kT/e) to red (−20 kT/e). The dashed lines represent the cut plane.
