TRAIL generally induces cell death through apoptosis (top left panel). Loss of MAP3K7 switches cell fate from apoptosis to necroptosis in response to TRAIL (top middle panel). In this context, necrosome components (RIPK1 and RIPK3) are recruited to the autophagy machinery via p62, leading to phosphorylation of MLKL and execution of necroptosis (lower panel). When necroptosis is inhibited by a RIPK3 inhibitor, (RIPK3i, GSK872) or by RIPK3, MLKL, or p62 knockdown, MAP3K7−/− cells undergo apoptosis in response to TRAIL (top right panel).
