Figure 1 Photographs (×400) of rabbit oocytes showing different germinal vesicle chromatin configurations. Photographs A and A', B and B', C and C', D and D', E and E', and F and F' are the same oocytes observed with phase contrast and fluorescence, respectively, after Hoechst staining. In the nonsurrounded nucleolus germinal vesicle, which was found only in the primordial follicles (with a single layer of flattened follicular cells), the diffuse filamentous chromatin was distributed all over the nuclear area and the nucleoli (arrows) were not surrounded with condensed chromatin (A, A'). In the netlike germinal vesicle, which occurred in both the primary follicles (with one or more layers of cuboidal follicular cells) (B, B') and the early antral follicles (C, C'), chromatin condensed into a netlike structure and the nucleoli (arrows) were surrounded with condensed chromatin. In the loosely condensed germinal vesicle (D, D'), the chromatin formed small clumps irregular in shape that either distributed over the nucleoplasm or surrounded the nucleoli (arrows). In the tightly condensed germinal vesicle (E, E'), chromatin further condensed into larger clumps with regular edges that were distributed throughout the nucleoplasm or surrounded the nucleoli (arrows). In the singly condensed pattern (F, F'), the chromatin condensed into a single large clump and the nucleoli disappeared completely. Figure 2 Detection of bromouridine incorporation in netlike (A, A'), loosely condensed (B, B') and tightly condensed (C, C') rabbit germinal vesicle oocytes. (A, B, C) Hoechst staining; (A', B', C') immunofluorescent detection of bromouridine incorporated into nascent RNA. Photograph magnification, ×400.
