ZO-1 or ZO-2 is important for clustering of claudins and occludin, resulting in the formation of tight junctional strands. The role of the other scaffolding proteins (ZO-3/MAGI/MUP1) is less clear. The ZOs and cingulin can provide a direct link to the actin cytoskeleton.
