3 Figure 2 Unstimulated (dashed line) and CR3/TNFα-stimulated (solid line) release of lactoferrin by adhering PMN of one study participant throughout the active study phase (0 minutes to 48hours). Study medication was administered over 2hours (shaded area, 0 to 120 minutes), blood for the isolation of PMN and subsequent analysis of lactoferrin release was sampled at t = 0, 30, 120, 180 minutes, and 48hours. In vitro lactoferrin release was estimated over 2hours in intervals of 30 minutes. For further analysis, the area under each curve was calculated using the trapezoid rule [29]. Abbreviations are: TFNα, tumor necrosis factor alpha; PMN, polymorphonuclear leukocytes. 4 Figure 3 Median number of killed E. coli/PMN (A), median killed E. coli in % of total number of phagocytosed E. coli /PMN (B), and median number of viable E. coli/PMN (C) at t = 0, 30, 120, 180 minutes, and 48hours after parenteral administration of 300mg of iron sucrose (•) or placebo (○). Bars indicate the corresponding 95% confidence intervals. Inserts indicate the level of significance of the treatment-time interaction. PMN, polymorphonuclear leukocytes. 5 Figure 4 Median released lactoferrin by PMN in the absence (A) or in the presence of concurrent stimulation with 20ng/mL TNFα (B) at t = 0, 30, 120, 180 minutes, and 48hours after parenteral administration of 300mg of iron sucrose (•) or placebo (○). Bars indicate the corresponding 95% confidence intervals. Lactoferrin is expressed as the median area under the curve (see Figure 2) divided by duration of the assay (120 minutes), yielding ng lactoferrin/105 PMN/minute. Inserts indicate the level of significance of the treatment-time interaction. PMN, polymorphonuclear leukocytes; TFNα, tumor necrosis factor alpha.
