Schematic Indicating Multiple Roles of cIAPs in Regulating Signaling Pathways
(A) cIAPs regulate multiple signaling pathways by building K63-linked ubiquitin chains. Yang et al. show that they also perform this same function in CARMA-Bcl10-MALT1 signaling. Red text: key molecules and concepts described in Yang et al. cIAPs are also required to inhibit the activation of non-canonical NF-κB by targeting NIK for proteasomal degradation by the addition of K48-linked ubiquitin chains.
(B) Birinapant promotes auto-ubiquitylation and proteosomal degradation of cIAPs, thereby preventing the B cell receptor signaling survival and activating non-canonical NF-κB. A role for TNF-induced death was not explored in Yang et al. but may contribute to cell death.
