Effects of phospholipids on the in vitro RNA-binding activity of the TBSV p33C replication protein. (A) EMSA analysis of the in vitro binding of purified TBSV p33C (p33 carrying the soluble C-terminal half) to 32P-labeled wild-type RII(+)-SL template (∼0.1 pmol). The assay contained the shown phospholipids added in comparable amounts (70 μM). The free or the RdRp-bound ssRNA was separated on nondenaturing 5% acrylamide gels. Each experiment was repeated at least two times. (B) Similar EMSA analysis as in panel A, except the p33C was preincubated with the shown phospholipids (70 μM each) for 5 min prior to the addition of the 32P-labeled RII(+)-SL RNA probe.
