Figure 1 A High-Throughput Method to Determine the Input Functions within a Gene Regulatory Network An existing E. coli fluorescent reporter library is used to provide individual clones, each with a promoter of interest driving green fluorescent protein (GFP) expression. The fluorescence emitted from the clone when grown in a range of cAMP and sugar (e.g., galactose) concentrations represents the transcriptional output of the promoter in response to the inputs. The input functions can be related to the known circuitry of the gene network and used to build an input-function map that describes how the cAMP concentration affects each gene directly and how the sugar concentration affects each gene through the network.
