Figure 1 Pol II Initiation, Pausing, and Release
Recruitment of Pol II by GTFs results in the formation of the PIC. After phosphorylation of the serine 5 of the CTD (pS5) by CDK7, Pol II enters into pause 30–50 nt downstream from the transcription start site (+1). Pol II pausing occurs through the recruitment of DSIF and NELF and is facilitated by the +1 nucleosome. CDK9, as part of P-TEFb, mediates the release of paused Pol II by the phosphorylation of DSIF, NELF, and the serine 2 of the CTD (pS2). Phosphorylation of NELF induces its release from Pol II.
