2 Figure 1 Ablation of dendritic cells in spleens and endometriosis lesions. Representative images of single cell suspensions derived from spleens (A) and induced endometriosis lesions (B) of the four groups of mice (i.e. WT mice treated with vehicle or DT and CD11c+ DTR/GFP transgenic mice treated with vehicle or DT), stained with CD11c allophycocyanin antibody and analysed for allophycocyanin and GFP fluorescence. Note that, in both spleens and endometriosis lesions, treatment of WT mice with DT did not affect the dentritic cell population, while treating CD11c+ DTR/GFP transgenic mice with DT led to a prominent ablation of dendritic cells. DT=diphtheria toxin; DTR=diphtheria toxin receptor; GFP=green-fluorescence protein; WT=wild-type. Reproductive BioMedicine Online  , DOI: ( /j.rbmo ) Copyright © 2014 Reproductive Healthcare Ltd. 3 Figure 2 Reduced growth of endometriosis lesions upon dentritic cell ablation. (A) Representative images of day-10 endometriosis lesions in CD11c+ DTR/GFP Tg mice treated with DT (top) versus vehicle control (bottom). (B) Bar histogram indicating the lesion areas of endometriosis lesions of the different experimental groups (n=5, 5, 4 and 4 mice in groups I, II, III and IV respectively, four lesions in each mouse); Values are mean±SD; the reduced size of lesions in DT-treated CD11c+ DTR/GFP Tg mice was statistically significant (P<0.0001). (C) Haematoxylin and eosin staining of histological sections of representative day-10 endometriosis lesions in CD11c+ DTR/GFP Tg mice treated with DT (group III; top) versus WT mice treated with DT (group II; middle) and CD11c+ DTR/GFP Tg mice treated with vehicle control (group IV; bottom); images on the right are magnified regions marked by a rectangle in images on the left; bars=1mm (left) and 0.1mm (right). DT=diphtheria toxin; DTR=diphtheria toxin receptor; GFP=green-fluorescence protein; WT=wild-type. Reproductive BioMedicine Online  , DOI: ( /j.rbmo ) Copyright © 2014 Reproductive Healthcare Ltd.
