Figure 1 P12+platelet-derived growth factor (PDGF) promoted adult human dermal fibroblast (AHDF) survival under nutrient deprivation. AHDFs were treated with PDGF±P12 in Hank’s balanced salt buffer (HBSS) for 72hours. Unless otherwise indicated, all experiments were performed in HBSS with 1nM PDGF and 10μM P12. Figure 2 Alexa488-P12 was endocytosed by an energy-dependent pathway. All experiments were performed with 10% fetal bovine serum in Dulbecco’s modified Eagle’s medium (DMEM). (a) Adult human dermal fibroblasts (AHDF) were treated with 1μM of Alexa488-P12 for 1hour at 37°C then cross-section imaged at 0.74μm. Bar=10μm. (b) AHDF were treated with 0.1,1, 10, or 50μM of Alexa488-P12 or 1μM Alexa Fluor-488 for 1hour at 37°C, then trypsinized and analyzed by flow cytometry. Figure 3 P12 increased platelet-derived growth factor BB (PDGF-BB)/PDGF receptor- β (PDGFR-β) internalization into macropinosomes. (a) MCF-7 cells expressing green fluorescent protein (GFP)-PDGFR-β were treated with TR-dextran and 1nM PDGF-BB±10μM P12 for 15minutes in Hank’s balanced salt buffer (HBSS), and then analyzed by confocal microscope. For dynamin inhibition, cells were pre-incubated with 80μM dynasore for 1hour in serum-free-Dulbecco’s modified Eagle’s medium (DMEM; see Supplementary Figure S6 online for clathrin heavy chain knockdown (CHC KD). Bar=10μm. (b) Colocalization of red and green signal was used to indicate the degree of colocalization between PDGFR-β and TR-dextran (threshold for both channels were set to 70). The PDGFR-β internalized during treatment was separated (see Materials and Methods section) and probed with antibodies against p-PDGFR-β (Tyr751) and p-extracellular signal–regulated kinases 1/2 (ERK1/2). Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was used as loading control. (b) AHDF cells were treated with 1nM PDGF-BB±10μM P12 in HBSS.
