FIGURE 4 Western blot analysis of E3.5 embryo ex vivo, cultured for 24 hours in Ham's F10+BSA and KSOM+AA. Mouse embryo isolated on E3.5 tested for p38MAPK all forms and phosphorylation, SAPK/JNK all forms, and phosphorylation expression by Western blot analysis (described in the Materials and Methods section) showed correct sized bands at 43 kDa for p38MAPK and 46 and 54 kDa for SAPK/JNK. Both SAPK/JNK and p38MAPK phosphorylation was greatly increased when embryos were cultured for 24 hours in Ham's F10, compared with KSOM+AA. Ham's F10 vs. KSOM+AA: SAPK/JNK phospho/SAPK/JNK all forms = 1.45, 0.78; p38MAPK phospho/p38MAPK all forms = 1.15, Note that 80–110 embryos were used in all groups, but some loss of embryo lysate may occur (for example, Ham's F10+BSA group for p38MAPK).
