Apoptosis Is Involved in Acute Cardiac Allograft Rejection in Rats
Yoshihiko Kageyama, MD, Xiao-Kang Li, MD, Seiichi Suzuki, MD, Hidetoshi Suzuki, MD, Kazuya Suzuki, MD, Teruhisa Kazui, MD, Yukio Harada, MD  The Annals of Thoracic Surgery  Volume 65, Issue 6, Pages (June 1998) DOI: /S (98)

Fig 1 Hematoxylin and eosin staining of left ventricular (A) isograft and (B) allograft sections at 5 days after transplantation. Infiltrating cells in the allografts showed an increase in number when compared with the syngeneic grafts. The Annals of Thoracic Surgery  , DOI: ( /S (98) )

Fig 2 Detection of DNA fragmentation in the (A–C) isograft and (D–F) allograft left ventricle at (A, D) 1, 3 (B, E), and 5 (C, F) days after transplantation. The allografts display positively stained myocytes with morphologic features of apoptosis, including dense nuclear margination. The Annals of Thoracic Surgery  , DOI: ( /S (98) )

Fig 3 Histogram of the apoptotic index (AI) values. Apoptotic cells were detected using in situ DNA nick-end labeling. Values were calculated for randomly chosen appropriate fields at a magnification of 200× using FUJIX, HC-2000, and Mac SCOPE/PPC software. The AI of allografts (Allo) was increased in comparison with that of isografts (Iso). (∗p = 0.015, ∗∗p = ) The Annals of Thoracic Surgery  , DOI: ( /S (98) )

Fig 4 Messenger RNA expression for Fas, Fas ligand (Fas-L), granzyme B (Gr. B), and perforin in syngeneic (Syn) and allogeneic (Allo) grafts. Fas transcripts were constitutively expressed in both the syngeneic and allogeneic grafts, and the level of expression was not dramatically altered after grafting. Fas ligand messenger RNA was undetectable in syngeneic grafts, but was upregulated in rejecting allografts. Expression of granzyme B and perforin messenger RNA was also upregulated in rejecting allografts. The Annals of Thoracic Surgery  , DOI: ( /S (98) )

Fig 5 Ultrastructural observation of the left ventricle from an allograft 3 days after transplantation. The characteristic features of apoptosis, such as chromatin condensation (bold arrow), formation of apoptotic bodies (fine arrow), and absence of surface microvilli, can be seen. The Annals of Thoracic Surgery  , DOI: ( /S (98) )
