Losartan-sensitive renal damage caused by chronic NOS inhibition does not involve increased renal angiotensin II concentrations  A. Marjan G. 2 Figure 1 Systolic blood pressure (A), glomerular filtration rate (B), proteinuria (C), renal Ang II (D), and blood Ang II (E) in control rats and rats treated with Nω-nitro- L-arginine (L-NNA) during 4, 7, 14, and 21 days.*P < 0.05 vs. control (day 0). 3 Figure 2 Representative light microscopic features of renal morphological changes after 4, 7, 14, and 21 days of treatment with L-NNA. (A) After four days of treatment with L-NNA, only very slight vascular, specifically endothelial injury, was observed, with sticking of some mononuclear cells to endothelial cells. (B) After seven days of treatment with L-NNA, mild to moderate vascular injury with pronounced mononuclear cell sticking to the endothelium, and infiltration of intima and media could be observed; the periadventitial space was also moderately infiltrated by mononuclear cells. (C) After 14 days of L-NNA treatment, moderate vascular injury with sticking of mononuclear cells to prominent vacuolated endothelial cells that were partly necrotic was observed; also, some vascular smooth muscle cells near the adventitia were necrotic and surrounded by mononuclear cells. (D) After 21 days of treatment with L-NNA, focal chronic tubular interstitial damage with collapse of tubular lumina, irregular vacuolation of tubular epithelia, and some infiltration of the interstitium by mononuclear cells could be seen. The glomerulus shows slight ischemic damage with condensation of mesangium and collapse of some capillaries (period acid-Schiff stain). 4 Figure 3 (A) Systolic blood pressure in control rats (squares) and rats treated for 21 days with L-NNA (circles), losartan (diamonds), or L-NNA + losartan (triangles).*P < 0.05 vs. control; #P < 0.05 vs. losartan. (B) Proteinuria in control rats (triangles) and rats treated for 21 days with L-NNA (circles), losartan (diamonds), or L-NNA + losartan (triangles). *P < 0.05 vs. control. 5 Figure 4 Representative light microscopic feature of renal morphology after 21 days of combined administration of L-NNA + losartan. No renal morphological changes were observed. The figure shows a regular glomerulus with typical mesangium and an arteriole with an increase in renin producing cells (bright red intracellular droplets). The interstitium is not edematous; proximal and distal tubules are regular (periodic acid-Schiff stain). Publication of this figure in color was made possible by a grant from Merck Sharp & Dohme BV, Haarlem, The Netherlands.
