3 Figure 2 Western blot probed with anti-bFGF antibody shows bFGF expression in the culture medium of pCAcchbFGFcs-treated rat fibroblasts. Control, control medium; bFGF, culture medium of pCAcchbFGFcs-treated cells; arrows, bands of bFGF. 4 Figure 3 Western blot probed with anti-bFGF antibody showing bFGF expression in rat dorsal muscle at 4 days after injection of pCAcchbFGFcs followed by electroporation. NM, normal muscle of rat back; LacZ, pCAZ3-treated dorsal muscle; bFGF, pCAcchbFGFcs-treated dorsal muscle; arrows, bands of bFGF. 5 Figure 4 Evaluation of flap necrosis (A–E), microangiographic findings (F–J) and blood vessel density (K) at 7 days after flap elevation. Photographs show dorsal skin flaps of rats from LacZ-E− group (A), FGF-E− group (B), LacZ-E+ group (C) and FGF-E+ group (D), and the area of flap necrosis was measured (E). Arrow head indicates boundary line between necrotic and viable flap. Angiographs show flap vasculature of rats from LacZ-E− group (F), FGF-E− group (G), LacZ-E+ group (H) and FGF-E+ group (I). The vasculature in the ischemic skin flap was quantified as the ratio of opacity in the distal (cranial) third of the flap to that in middle third of the flap (J). Endothelial cells were immunostained in each cross section of the skin flap, and the number of vessels in the panniculus carnosus (distal third) was counted (K). Values are shown as mean±SD (*P<0.01, versus other three groups).
