The mammalian circadian system is organized hierarchically, whereby light-entrainable brain pacemaker neurons in turn synchronize peripheral tissue clocks, giving rise to rhythmic physiologic outputs important in glucose homeostasis, feeding rhythms, and energy balance. Illustrated is an in vitro screen to identify modulators of the molecular clock which are present in both brain and peripheral cells (core clock components are illustrated: CLOCK, circadian locomotor output cycles kaput; BMAL1, brain Arnt-like protein 1; CRY, cryptochrome; PER, period; REV-ERB, reverse strand of the Erb B receptor; ROR, retinoic acid receptor-like receptor; FBXL3 and FBXL21, F-box and leucine-rich repeat protein 3 and 21; CK1ε/δ, caseine kinase 1 ε/δ).
