(A) Survival of B6-WT or B6-STING-/- recipients after 11.5 Gy irradiation on day-1 and transplant with 7 × 106 BM cells and ~4 × 106 LN cells (normalized to 1.7 × 106 T cells) or (B) after 10.5 Gy irradiation on day 0 and transplant with 7 × 106 BM cells and ~4 × 106 LN cells (normalized to 1.8 × 106 T cells) from BALB/c donors. (C) Fold induction of IFNβ, TNFα and IL-6 RNA from B6-WT or B6-STING-/- recipient colon tissue 48 hrs. after 8.5 Gy irradiation on day 0 and transplant with 10 × 106 BM cells and ~2.5 × 106 spleen cells (normalized to 8 × 105 T cells) from LP/J donors. (D) GVHD clinical score of B6-WT or B6-STING-/- recipients after 10.5 Gy irradiation on day 0 and transplant with 7 × 106 BM cells and ~24 × 106 spleen + peripheral LN cells (normalized to 2 × 106 CD8+ T cells) from C3H.SW donors. (E) Survival of B6-WT and B6-STING-/- recipients transplant as in (D) with IP injection of DMXAA (25 mg/kg) or vehicle (5% NaHCO3) at the time of irradiation. All data show mean + SEM.
