2 Fig. 1 General structural organization of calpains. Catalytic subunits have four domains (DI–DIV) and regulatory subunits have two domains (DV and DVI), respectively. DI is autolysed during calpain activation by calcium. DII is the conserved protease domain, which is sub-divided into the subdomains such as IIa and IIb, which form a signal domain during calcium binding. DIII consisting C2 domains, which is participates in structural changes during calcium binding. The carboxy-terminal domains DIV and DVI contain five EF hands and the fifth EF hand involves the subunits dimerization. This figure adopted from Harris et al. (2004).
