2 Figure 1 RNA interference using DNA vector–based short interfering (si) RNA technology. (a) Predicted secondary transcripts in hairpin structures used to target OB-R or used as control. (b) Schematic drawing of the expression vector. (c) HEC1A cells (I), OB-R short hairpin (sh) RNA transfected cells (II), and scrambled shRNA transfected cells (III). GFP fluorescence is present in both OB-R (IV) and scrambled (V) shRNA transfected cells. Cervero. Embryonic adhesion not affected by OB-R silencing. Fertil Steril 2007. 3 Figure 2 Specific silencing of OB-R expression. (a) Quantitative mRNA analysis of OB-R in HEC1A nontransfected cells (H), OB-R short hairpin (sh) RNA transfected cells (H-OBR), and scrambled shRNA transfected cells (H-SCR). Data are expressed in arbitrary units and represented as mean ± SD. Statistical differences are present in the OB-R expression between OB-R shRNA cells and in nontransfected cells (P≤.0001) or cells transfected with the scrambled shRNA sequence (P≤.0001). (b) Western blot analysis of OB-R expression in nontransfected cells (H) or cells transfected with OB-R (H-OBR) and scrambled shRNA (H-SCR). (c) Immunocytochemistry analysis of OB-R expression in control cells (I), scrambled transfected cells (III), and cells transfected with OB-R shRNA vector (V). In control experiments, immunostaining was not present when the primary antibody was omitted (II, IV, VI). (d) OAS1 induction assessment in control cells (H) and transfected cells (H-OBR and H-SCR). No statistical differences were found in OAS1 expression between the three groups (P≥.4). Cervero. Embryonic adhesion not affected by OB-R silencing. Fertil Steril 2007. 4 Figure 3 Embryonic adhesion assays. (a) Scheme of adhesion model composed by mouse blastocysts and HEC1A endometrial cells. (b) A total of 420 murine blastocysts were cocultured with nontransfected cells (H) and cells transfected with OB-R short hairpin (sh) RNA (H-OBR) and scrambled shRNA (H-SCR) in four different experiments. Adhesion rates were 94 ± 2.8%, 92 ± 9.8% and 94 ± 4.6% for nontransfected, OB-R shRNA transfected cells, and scrambled shRNA transfected, respectively. Student t test did not show statistical differences (P≥.6). Cervero. Embryonic adhesion not affected by OB-R silencing. Fertil Steril 2007.
