Figure 1 Leptin and Ob-R expression in the human endometrium. (A) Leptin and Ob-R mRNA expressions. Total RNA was extracted from endometria biopsies of fertile and infertile women and then analyzed by quantitative RT-PCR. The values are the mean ± SEM and are expressed as percentages of the control value (fertile subjects). ∗P<.05; ns = nonsignificant, unpaired t test. (B) Leptin protein expression. Endometrial explants were fixed, paraffin-embedded, and stained for anti-leptin antibody as described in Materials and Methods. This figure shows one representative experiment among five separate experiments. (C) Ob-R protein expression. Cell lysates (50 μg) were extracted from endometria biopsies of fertile (CONT) and infertile (IF) women and then subjected to Western blot analysis using anti–Ob-R or anti–β-actin antibody, as described in Materials and Methods. This figure shows one representative experiment among 8–10 separate experiments. The values are the mean ± SEM and are expressed as percentages of the control value (fertile subjects). ∗P<.05; ns = nonsignificant, unpaired t test.
