2 Figure 1 Multiple Roles of the Nuclear Localization Signal in Cell Function and Disease A generalized cartoon of the multidomain structure of a typical RNA binding protein (RBP), highlighting LC, RGG, RRM and NLS (PY-NLS in case of FUS) domains. Cation-π interactions can contribute to phase separation as shown for FUS. Upon stress, RBPs can relocate to stress granules (SGs). Amyloid fibers can emerge by assembly of RBPs. Nuclear import receptors (NIRs) employ different approaches to remove proteins from condensates such as SGs, including competition with RNA binding and inhibiting/chaperoning aberrant phase transitions like those implicated in ALS and FTD. Conventionally, NIRs ferry FUS and other RBPs through nuclear pore complexes. Cell  , DOI: ( /j.cell ) Copyright © 2018 Elsevier Inc. Terms and Conditions
