(A) IRG1/CAD is induced upon inflammation and converts cis-aconitate to itaconate. Itaconate has direct antimicrobial effects on both intracellular bacteria and extracellular bacteria. Additionally, itaconate can inhibit succinate dehydrogenase activity, leading to decreased ROS production and inflammation. (B) IRG1/CAD decarboxylates cis-aconitate to generate itaconate.
