Figure 1 Agrin-DGC-Yap Signaling Axis Controls Cardiomyocyte Maturation and Cardiac Regeneration (A) Neonatal expression of the ECM protein Agrin by the endothelial cells maintains the undifferentiated state of CMs, allowing CM proliferation and cardiac regeneration. At the end of the regenerative time frame (P7), Agrin is downregulated and CMs go through maturation, resulting in greatly reduced regenerative capacity. (B) Binding of Agrin to the DGC complex induces the release and nuclear localization of the Hippo effector Yap in the neonatal heart. (C) In the adult heart, coinciding with the CM maturation, other ECM components such as laminin may replace Agrin, causing the maturation of DGC and tethering of Yap in the cytoplasm.
