The cartoon illustrates the hypothesis that the phosphotyrosine stabilizes the active conformation of the catalytic domain (cat). (B) DYRKs promote cell-cycle exit. Phosphorylation by DYRKs promotes proteasomal degradation of cyclin D1 and c-Myc, which drive S-phase entry. Antiproliferative CDK inhibitors are transcriptionally upregulated (p21Cip1) or stabilized (p27Kip1). (C) Proposed role of DYRK1A in Alzheimer’s disease. DYRK-mediated hyperphosphorylation of the tau protein induces dissociation of tau from the microtubules, destabilization of microtubules and aggregation of tau in paired helical filaments, which cause neurotoxicity.
