-Tests that light & chloroplasts are needed for light reactions to occur DPIP replaces NADP+ as the final electron acceptor DPIP becomes when reduced (gaining electrons) Color change can be measured with spectrophotometer Experiment – write this summary down!! Set wavelength to 605 – check spec filter Prepare cuvettes from Table 4.3 (Don’t add chloroplasts….yet) Blank spec – read instructions next to wavelength knob Add chloroplasts & invert 2X to mix Place in spec & record for time 0 minutes Place in front of light Record at 5 minute intervals (get chloroplasts on ice please) Lab write-up - due WEDNESDAY just what is on the sheet….no pre-lab, purpose, procedure
