On the left, the structure of JAK1 bound to box1 from the IFNλ receptor (PDB: 5IXD (Ferrao et al., 2016)) is shown overlaid with the box2 motif from the IFNα receptor (PDB: 4PO6 (Wallweber et al., 2014)). The PxxLxF motif within box1 identified by Ferrao et al. (2016) is shown with the sidechains colored white. Although box1 and box2 occupy non-overlapping, nearly continuous binding sites, they have not yet both been observed in the one structure. On the right is a schematic of a full-length JAK/Receptor interaction. For clarity, only one receptor chain and associated JAK is shown.
