Figure 1 Generalized mode of action of SOCS proteins. The SH2 domain binds to phosphorylated tyrosines on JAKs or on the receptor and positions the N-terminal domain to interact with and inhibit JAK kinase activity. The SOCS box binds elongin C and recruits an E3 ubiquitin ligase-like complex that targets bound proteins to proteolytic degradation. These actions terminate the cytokine activation cycle.
