Generalized mode of action of SOCS proteins. The SH2 domain binds to phosphorylated tyrosines on JAKs or on the receptor and positions the N-terminal domain to interact with and inhibit JAK kinase activity. The SOCS box binds elongin C and recruits an E3 ubiquitin ligase-like complex that targets bound proteins to proteolytic degradation. These actions terminate the cytokine activation cycle. Several other binding interactions of SOCS proteins have been described, but their significance is not yet clear
