2. Figure 1 Activity Analyses of GR24 Stereoisomers.
(A) Structures of 5-deoxystrigol, GR24 (rac-GR24, racemic mixture of GR245DS and GR24ent-5DS) and GR24 stereoisomers. S and R show the configuration of 3a-C, 8b-C in the C ring and 2′-C in the D ring.
(B) Retained content of GR24 stereoisomers in solution. GR24 stereoisomers were incubated in the absence or presence of AtD14. The amounts of GR24 stereoisomers remaining in solution were measured by HPLC-MS/MS and scaled to a relative percentage compared with GR24 stereoisomers in the solution containing no protein component. Data are represented as means ± SEM (n = 3 biological replicates). a, b, c indicate the statistically significant analysis for results without D14; parentheses have been added to the letters for results with D14; P < 0.05 (ANOVA).
(C) Pull-down assay using His-MAX2 (as the bait) and GST-AtD14 in the absence or presence of 10 μM GR24 or GR24 stereoisomers; or 20 μM GR24ent-4DO. GST-AtD14 was detected by anti-GST antibody (top panel), and the amount of His-MAX2 was shown by staining the PVDF membrane (bottom panel).
(D) Pull-down assay using Flag-SMXL7 (as the bait) and GST-AtD14 in the absence or presence of 10 μM GR24 or GR24 stereoisomers; or 20 μM GR24ent-4DO. The amount of Flag-SMXL7 was shown by staining the PVDF membrane (bottom panel).
(E) Relative hypocotyl length of 7-day-old Col-0 seedlings growing on MS medium with different concentrations of GR24 or GR24 stereoisomers. Data are represented as means ± SEM (n = 30).
