Colors designate HFE domains encoded by individual exons (cyto, cyotoplasmic; tm, transmembrane). HFE requires interaction with β2-microglobulin (β2M) for normal cell surface localization. While transferrin receptor 1 (TfR1) interacts with the HFE α1 and α2 domains, TfR2 interacts with the HFE α3 domain. Regulation of hepcidin by holotransferrin requires HFE sequences in the α3 (and cytoplasmic) domains and interaction with transferrin receptor 2.
