Amplification of total RNA from endometriosis lesions for epithelial neutrophil-activating peptide 78. Two independent endometriotic lesions (lanes 1 and 2) expressed the expected 162–base pair product, whereas no bands were identified in the control lane (NT2-N cells) (lane 3). Peritoneal macrophages also clearly expressed the expected 162–base pair amplicon (lane 4).

(A) In situ hybridization of an endometriosis lesion with 35S-labeled RNA antisense probe specific for ENA-78 demonstrates that ectopic endometrial glandular cells express this chemokine. (B) No signal is visible in the negative control hybridized with a sense probe. (C) Scattered cells within the endometriotic stroma also express ENA-78 transcripts. (D) Colocalization with CD68 antibodies (magenta staining) indicate that some of these stromal cells are tissue macrophages, whereas the remaining mesenchymal cells appear to be of fibroblastic origin.
