2 Fig 1 Proposed slit diaphragm morphology and structure. In the original paper by Rodewald and Karnovsky,4 the slit diaphragm was shown to be a zipper-like structure by electron micrography (A) and was postulated to be approximately 39 nm in width with a central filament and flanking crossbridges with intervening 4 × 14 nm pores (B). As proposed by Tryggvasson,23 the new molecular model for the slit diaphragm consists of nephrin molecules from opposing foot processes interdigitating their Ig repeats 1 through 6, thus forming the central zipper-like structure observed in the electron microscopy studies (C).
